Product Specific Information

The eBioC9B7W monoclonal antibody recognizes mouse CD223 (LAG-3, LAG3) protein expressed by activated α/β-TCR-bearing T cells, subsets of γ/δ-TCR-bearing T cells, and NK cells.
CD223 is a 70 kDa type I transmembrane protein structurally similar to CD4. A soluble form of CD223 can be generated by proteolytic cleavage, producing a 54 kDa extracellular fragment and a 16 kDa intracellular fragment.

CD223 binds to MHC class II with higher affinity than CD4, contributing to negative regulation of T-cell activation and proliferation. It is also expressed by CD4+CD25+ regulatory T cells and may play a role in their regulatory function.

Applications Reported: Flow cytometry, immunoprecipitation, ELISA.
For functional assays, use Functional Grade purified eBioC9B7W (C9B7W) Product #16-2231-85.

Applications Tested:

  • Flow cytometric analysis of anti-CD3 and anti-CD28-activated mouse splenocytes (≤0.5 µg/test, 100 µL final volume).
  • ELISA analysis using supernatant from mouse CD223-transfected 3A9 cells (2 µg/mL for capture; detection with polyclonal anti-mouse CD223, Product #24-2232).

Target Information

LAG-3 is a 70 kDa surface glycoprotein of the Ig superfamily with homology to CD4. It binds MHC class II with higher affinity than CD4 and negatively regulates T cell activation and homeostatic proliferation.
LAG-3 is expressed on activated T cells (including regulatory T cells) and NK cells; CD8+ T cells typically express higher levels than CD4+ T cells. Coexpression of LAG-3 and CD49b identifies Type 1 regulatory T cells (Tr1) in both human and mouse.
