Product Specific Information

The LWC06 antibody was generated by immunization with the recombinant extracellular region of mouse CIRE/DC-SIGN (CD209).
CIRE/DC-SIGN is a 33 kDa type II transmembrane C-type lectin protein expressed on CD8α⁻ dendritic cells and plasmacytoid predendritic cells, and is the closest homologue of human DC-SIGN.
It binds mannose and other carbohydrates in a calcium-dependent manner and may play a role in T-dendritic cell interactions through ICAM family binding.

CIRE/DC-SIGN expression varies by dendritic cell subpopulation and activation state. It is down-regulated in GM-CSF–supplemented spleen-derived dendritic cell cultures.
Unlike human DC-SIGN, which is dendritic cell–specific, CIRE/DC-SIGN mRNA is also detected in B cells.

The LWC06 monoclonal antibody does not cross-react with SIGNR1, SIGNR2, SIGNR3, or SIGNR4.
CD209 protein is sensitive to collagenase treatment.

Applications Reported: Immunoprecipitation (IP) and Western blot (WB).
Applications Tested: Tested in CIRE-transfected CHO cells by IP and WB using reciprocal detection with another CIRE-specific antibody (clone 5H10, Product #14-2091).

Target Information

This gene encodes a transmembrane receptor often referred to as DC-SIGN, expressed on dendritic cells and macrophages.
It functions in innate immunity by recognizing a wide range of pathogens (parasites to viruses).
The protein has three main domains:

  • N-terminal transmembrane domain
  • Tandem-repeat neck domain
  • C-type lectin carbohydrate recognition domain

The extracellular region mediates pathogen recognition and cell adhesion by binding carbohydrate ligands.
The neck domain supports homo-oligomerization, enabling high-avidity binding to multivalent ligands.
Variation in the number of 23-amino-acid repeats in the neck region affects ligand-binding ability.
This gene is closely related to L-SIGN (GeneID 10332) but differs in ligand-binding properties and tissue distribution.
Alternative splicing results in multiple variants.
