The eBioL31 monoclonal antibody reacts with mouse CD207 (Langerin), expressed in a subset of dendritic cells known as Langerhans cells (LC). CD207 is a 48 kDa C-type lectin transmembrane protein involved in antigen recognition and uptake. LC in the epidermis reduce CD207 expression upon activation and migrate toward lymphatic vessels. Expression of CD207 correlates with Birbeck granule presence, and granule formation has been observed upon CD207 cDNA transfection into fibroblast lines.
Western blotting with eBioL31 detects a 48 kDa band in mouse ear epidermis lysates and CD207-transfected cells. Preliminary data suggest suitability for intracellular flow cytometric analysis using the Foxp3/Transcription Factor Staining Buffer Set (Product # 00-5523). The epitope is located in the extracellular CRD region, allowing detection of both surface and intracellular Langerin. Expression levels may vary among mouse strains.

Target Information
Langerin (CD207) is a type II membrane-associated C-type lectin expressed by Langerhans cells. It recognizes mannose residues via its carbohydrate recognition domain (CRD) and is localized both on the cell surface and intracellularly with Birbeck granules. Transfection of Langerin cDNA into fibroblasts induces membrane structures characteristic of Birbeck granules, indicating its role in membrane superimposition and BG formation.
