Target Information
PSD-93 (chapsyn-110) is a plasma membrane-associated protein found in synaptic junctions. It is uniquely expressed in Purkinje neuron cell bodies and dendrites. PSD-93 contains three PDZ domains (~90 amino acids each), one SH3 domain, and a carboxyl-terminal Guanylate Kinase homology (GuK) domain that is enzymatically inactive.
PDZ-domain interactions are thought to play roles in receptor and channel clustering, contributing to neuronal plasticity. PSD-93 participates in clustering of NMDA receptors and shaker-type potassium channels at the synaptic membrane. These proteins share a C-terminal T/SXV motif enabling PDZ-domain binding. Additionally, neuronal nitric oxide synthase (nNOS), which lacks this motif but has its own PDZ domain, associates with PSD-93 via pseudo-homotypic PDZ–PDZ interaction.
