Product Specific Information

This eyedio monoclonal antibody reacts with human indoleamine-2,3-dioxygenase (IDO, INDO, IDO1), an intracellular enzyme that catalyzes the degradation of tryptophan to kynurenines. IDO is expressed in macrophages, plasmacytoid dendritic cells, and various cell lines, and can be induced by IFN-gamma or other inflammatory stimuli. Expression of IDO in antigen-presenting cells and tumors is associated with immune suppression via tryptophan depletion and generation of metabolites that trigger T cell apoptosis and regulatory T cell induction.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and validated for flow cytometry of human peripheral blood cells using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00). Use at 5 µL (0.125 µg) per test (100 µL final volume). Cell count may vary between 10⁵–10⁸ cells/test.

Fluorochrome Information:
APC-eFluor 780 emits at 780 nm and is excited by the Red laser (633–647 nm). Verify instrument compatibility.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-57) for up to 3 days at 4°C in the dark with minimal brightness loss. Clone-specific performance should be empirically determined.

Target Information

IDO1 is a heme-containing enzyme that catalyzes oxidative cleavage of the indole ring of tryptophan, serotonin, and melatonin, producing kynurenines. It is expressed in many tissues, macrophages, and dendritic cells. IFN-induced IDO1 plays a key antimicrobial and immunoregulatory role by depleting tryptophan, inhibiting pathogen growth, and modulating immune responses during infection, pregnancy, autoimmunity, transplantation, and cancer.
