Product Specific Information
Description:
The 4S.B3 monoclonal antibody reacts with interferon-gamma (IFNγ), a 17 kDa cytokine produced by activated T and NK cells with antiviral and antiparasitic activity. It synergizes with TNFα to inhibit cell proliferation and modulate immune responses.
Applications Reported:
Validated for flow cytometric analysis.
Applications Tested:
Pre-titrated and tested by intracellular staining and flow cytometry of stimulated human peripheral blood cells. Recommended use: 5 µL (0.25 µg) per test in 100 µL final volume. Cell count: 10⁵–10⁸ cells/test (empirically determined).
Fluorophore Information:
APC-eFluor 780 emits at 780 nm, excited by a 633 nm red laser. Ensure instrument compatibility.
Light Sensitivity:
Protect from light to prevent photo-induced oxidation.
Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness or compensation.
Excitation/Emission:
633–647 nm / 780 nm (Red Laser)
Filtration:
0.2 µm post-manufacturing filtered.

IFN gamma (Interferon gamma, Type II interferon):
A macrophage activation and immune interferon produced mainly by T-lymphocytes and NK cells in response to antigens and cytokines.
- Dimeric glycoprotein (~45 kDa) consisting of two 146 amino acid subunits
- Functions: antiviral, antiproliferative, MHC induction, macrophage activation, B-cell immunoglobulin enhancement
- Species specificity: human IFNγ does not cross-react with mouse
- Regulated by IL2, FGF basic, EGF (upregulated); inhibited by vitamin D3 or DMN
- Mutations associated with aplastic anemia
