The polyclonal antibody reacts with mouse, rat, and human p21; it was raised against a peptide mapping to the carboxy terminus of human p21. Cell cycle progression is regulated by cyclins and Cdks, and can be inhibited by CDK inhibitors (CDIs) such as p21 (WAF1/Cip1). p21 inhibits cyclin/Cdk family activity and overexpression suppresses mammalian cell proliferation. Its expression is inducible by stress and enhanced by p53. Other CDIs include p27 (Kip1) and p16 (INK4A), both involved in cell cycle arrest and tumor suppression.

Target Information

Protein p21 is a tumor suppressor and CDK inhibitor activated by p53. It mediates cellular response to genotoxic stress and regulates cell cycle arrest for DNA repair. Nuclear p21 inhibits Cdk1 and Cdk2, blocking G1/S and G2/M transitions. Cytoplasmic p21 has anti-apoptotic roles by inhibiting caspase 3, ASK1, and JNK. p21 exhibits dual functions in carcinogenesis, acting as both tumor suppressor and oncogene depending on context.
