Product Specific Information

The polyclonal antibody reacts with mouse, rat, and human NFkB p50. It was raised against a peptide mapping to the carboxy terminus of human NFkB p50. Members of the rel/NFkB family of transcription factors regulate cellular responses such as growth, development, and inflammation. They share a structural motif known as the rel homology region (RHR), which mediates protein dimerization.

Complexes of p50 (NF-kB1) or p52 (NF-kB2) are generated through processing of p105 and p100 precursors, respectively. These complexes associate with Rel family members (p65, c-Rel, Rel B) to form homo- or heterodimers that bind distinct kB sites to regulate gene transcription.

In resting cells, NFkB is retained in the cytoplasm by inhibitory IkB proteins. Upon activation by cytokines, LPS, oxidative or mechanical stress, IkB is degraded, allowing NFkB to translocate to the nucleus and activate transcription of proinflammatory genes. NFkB activation has been linked to various pathological states such as cardiac ischemia-reperfusion injury and insulin-dependent diabetes mellitus.

Applications Reported: Purified anti-mo/hu/rat NFkB p50/p105 polyclonal antibody has been reported for use in immunoprecipitation, immunoblotting (WB), and immunohistochemical staining.
Applications Tested: Tested by immunoblotting (WB) at 1:400 dilution. Recommended to titrate for optimal performance.
Purity: >90% by SDS-PAGE.
Aggregation: <10% by HPLC.
Filtration: 0.2 µm post-manufacturing filtered.


Target Information

NF-kB is a nuclear transcription factor activated by various stimuli such as cytokines, UV radiation, stress, and bacterial or viral products. It regulates cellular events including growth, differentiation, proliferation, apoptosis, and inflammation.

NFKB1 (p50) is a 50 kDa subunit of NF-kB, synthesized as a p105 precursor containing the RHD region with nuclear localization, DNA-binding, and dimerization domains. p50 forms homodimers or heterodimers with p65, constituting functional NF-kB complexes. NFkB plays roles in AIDS, cancer metastasis, and rheumatoid arthritis pathophysiology.
