Description:
The eBio4E10 monoclonal antibody reacts with mouse JAML (junctional adhesion molecule-like protein), also known as AMICA. JAML is a glycosylated membrane protein with two Ig-like domains in its extracellular region. It is expressed on polymorphonuclear leukocytes (PMN) and participates in their transmigration through epithelial tight junctions via interactions with the epithelial coxsackie and adenovirus receptor (CAR). JAML is also found on lymphocyte subsets, including γδ T cell lines, and provides co-stimulatory signals for cell activation.

Applications Reported:
This antibody has been reported for use in flow cytometric analysis and immunoprecipitation.

Applications Tested:
Tested by flow cytometric analysis of mouse lysed whole blood. Recommended use ≤1 µg per test (100 µL final volume). Optimal concentration should be titrated for each assay. Cell number can range from 10⁵ to 10⁸ per test.

Target Information

AMICA is a transmembrane protein located on the plasma membrane of leukocytes, regulating their migration and activation through interaction with CXADR, a receptor on epithelial and endothelial cells. This interaction mediates activation of γδ T-cells, which play roles in tissue homeostasis and repair. Upon CXADR binding, JAML triggers downstream PI3-kinase and MAP kinase signaling, promoting proliferation and cytokine production in T-cells, which in turn stimulate epithelial tissue repair. It also facilitates leukocyte transmigration via adhesive interactions with CXADR.
