Product Specific Information
Description:
The 8.1.1 monoclonal antibody reacts with mouse podoplanin (T1a, gp38, aggrus), a 43 kDa transmembrane glycoprotein expressed in kidney glomerular epithelial cells (podocytes). Podoplanin is also found in epithelial and mesothelial cells such as intestinal epithelium, alveolar type I cells, podocytes, and visceral peritoneum mesothelium. It serves as a potent marker for lymphatic endothelium and is expressed by subcapsular epithelial cells of the murine thymus. Mice deficient in Podoplanin die at birth due to respiratory defects and congenital lymphedema caused by lymphatic pattern formation failure.
Applications Reported:
This eBio8.1.1 (8.1.1) antibody is suitable for flow cytometric analysis, immunoprecipitation, immunoblotting (WB), and immunohistochemical staining.
Applications Tested:
Tested by flow cytometric analysis of TE-71 cells. Use ≤0.5 µg per test (in 100 µL final volume). Optimal antibody titration is recommended for best assay performance.
Purity: Greater than 90%, as determined by SDS-PAGE.
Aggregation: Less than 10%, as determined by HPLC.
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
Podoplanin (PDPN, T1A, gp38, Aggrus) is a mucin-type 1 glycoprotein (40–43 kDa) expressed in tumors and normal cells, especially lymphatic epithelial cells and follicular dendritic cells. It localizes in stromal cells of peripheral lymphoid tissue and thymic epithelial cells. As a regulator of lymphatic endothelium, podoplanin helps maintain podocyte morphology and serves as a ligand for CLEC-2. Its expression correlates with tumor invasiveness and metastasis potential. Podoplanin is involved in cell migration, metastasis formation, and tumor invasion. Dysregulation of podoplanin is associated with diseases such as lymphangioma and subglottic neoplasm and has been proposed as a marker for lung injury.
