Product Specific Information

The eBioGHI/61 monoclonal antibody reacts with human CD163 (Haemoglobin Scavenger Receptor, M130), a macrophage scavenger receptor family member. CD163 is expressed on tissue macrophages, circulating monocytes, and possibly hematopoietic progenitor cells. It functions as a high-affinity receptor for the hemoglobin-haptoglobin complex, mediating clearance of hemoglobin from blood.

• Function: Binds hemoglobin-haptoglobin complex with high affinity; induces IL-6 and IL-10 production upon ligand binding.
• Regulation: Upregulated by IL-6 and IL-10; downregulated by LPS and TNF-α via shedding of soluble CD163.
• Applications Reported: Flow cytometric analysis.
• Applications Tested: Pre-titrated and tested on normal human peripheral blood cells.

  • Suitable cell range: 10⁵–10⁸ cells/test.
• Light Sensitivity: Protect from light due to photo-induced oxidation sensitivity.

Target Information

CD163 (M130 antigen, Ber-Mac3, Ki-M8, SM4) is a 130 kDa membrane glycoprotein, member of the scavenger receptor cysteine-rich superfamily, and receptor for the hemoglobin-haptoglobin complex. It protects tissues from oxidative damage and participates in iron recycling.

• Expressed on monocytes and macrophages, especially in late inflammation stages.
• Present on CD14⁺ and CD64⁺ monocytes; higher on CD16⁺ monocytes.
• Upregulated by IL-10, IL-6, dexamethasone; shedding induced by LPS and PMA.
• Binds hemoglobin/haptoglobin complexes in a calcium- and pH-dependent manner.
• Triggers intracellular signaling via tyrosine kinase, calcium mobilization, and IL-6/CSF1 secretion.
