Product Specific Information

This HP-3G10 monoclonal antibody reacts with CD161 (NKR-P1A), a C-type lectin superfamily member. CD161 is expressed on natural killer (NK) cells and various T cell subsets, including memory/effector CD4⁺ and CD8⁺ T cells, γδ TCR T cells, and a subset of CD3⁺ thymocytes. It is also found on human Th17 CD4⁺ T cells. The receptor’s function is not fully understood but may play a stimulatory role. Lectin-like transcript-1 (LLT1) is identified as its ligand.

Target Information

KLRB1 (CD161) is a type II membrane protein expressed on NK cells and a subset of CD3⁺ T cells. It forms a homodimeric surface antigen (40–44 kDa) and plays an inhibitory role in NK cell activity. Activation triggers sphingomyelinase/SMPD1 stimulation, increases ceramide levels, and induces AKT1/PKB and RPS6KA1/RSK1 kinase activity, promoting T cell proliferation.

KLRB1 binds to:

• Gal-α(1,3)Gal and N-acetyllactosamine epitopes
• CLEC2D/LLT1 ligand, inhibiting NK cell cytotoxicity and IFN-γ secretion

The protein includes an extracellular C-type lectin domain, transmembrane domain, and cytoplasmic domain.
