The M3.2D7 monoclonal antibody was generated against and reacts with mouse B7-H3 (CD276), a member of the B7 family of the Ig superfamily proteins. B7-H3 is expressed by antigen-presenting cells and developing bone during embryogenesis. Its ligand has not yet been identified, but it is thought to down-regulate T cell functions through an unknown counter receptor on T cells. M3.2D7 stains mouse B7-H3 transfected cells but not mouse spleen cells, and also stains human B7-H3 transfected cells.

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Tested by flow cytometric analysis of mB7-H3 transfected cells. Recommended ≤ 0.5 µg/test (100 µL final volume). Optimal titration is recommended depending on cell number (10⁵–10⁸ cells/test).

Target Information

CD276 (B7-H3) is a member of the B7 family of co-stimulatory molecules. It is a type I transmembrane protein that induces proliferation of CD4⁺ and CD8⁺ T cells, enhances cytotoxic T cell generation, and promotes interferon gamma production. Expression is inducible on dendritic cells and monocytes by inflammatory cytokines and is present in peripheral tissues such as heart, kidney, testis, and colon. In humans, CD276 exists as two isoforms due to gene duplication and alternative splicing. CD276 has potential therapeutic applications in regulating immune responses to cancer, particularly with anti-angiogenic therapy.
