Product Specific Information

The BTLA monoclonal antibody (clone MIH26) reacts with human B and T lymphocyte attenuator (BTLA).
BTLA is expressed by peripheral lymphocytes, splenic macrophages, developing B cells in the bone marrow, developing T cells in the thymus, and mature dendritic cells (but not immature bone marrow-derived dendritic cells).
BTLA acts as a negative regulator of activation and/or function of various hematopoietic cell types. It is reported to bind to B7-H4, although further studies are needed to confirm this interaction.

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Tested by flow cytometric analysis of normal human peripheral blood.
Recommended use ≤ 0.5 µg per test (a test = antibody amount for 100 µL cell sample). Cell number should be empirically determined (10⁵–10⁸ cells/test). Antibody titration is recommended for optimal performance.

Storage and Handling: Use in a sterile environment.
Filtration: 0.2 µm post-manufacturing filtered.
Purity: >90% (SDS-PAGE).

Target Information

BTLA (B and T-lymphocyte attenuator) is a member of the co-inhibitory receptors of the CD28 superfamily.
It functions as a lymphocyte inhibitory receptor that suppresses lymphocyte activation during immune responses.
BTLA is constitutively expressed on most CD4⁺ and CD8⁺ T cells, with expression decreasing upon T cell activation. It remains expressed on Th1 but not Th2 cells.
BTLA interacts with the TNFR superfamily member HVEM (herpesvirus entry mediator), forming an important pathway regulating lymphocyte activation and immune homeostasis.
