Target Information
PTEN is a tumor suppressor frequently mutated in various cancers.
It encodes a phosphatidylinositol-3,4,5-trisphosphate 3-phosphatase containing a tensin-like domain and a catalytic domain similar to dual-specificity protein tyrosine phosphatases.
Unlike most protein tyrosine phosphatases, PTEN preferentially dephosphorylates phosphoinositide substrates.
PTEN negatively regulates intracellular levels of phosphatidylinositol-3,4,5-trisphosphate and functions as a tumor suppressor by inhibiting the AKT/PKB signaling pathway.
It modulates the AKT-mTOR pathway, influencing neuron integration, positioning, dendritic development, and synapse formation during adult neurogenesis.
PTEN may also regulate insulin signaling and glucose metabolism in adipose tissue.
The nuclear monoubiquitinated form has greater apoptotic potential, while the cytoplasmic nonubiquitinated form exhibits reduced tumor suppressive ability.
