Target Information
Metadherin (Metastasis adhesion protein), also known as MTDH or LYRIC, is a novel protein that localizes with tight junction proteins ZO-1 and occludin in polarized epithelial cells. It is recruited during the maturation of the tight junction complex rather than acting as a structural component.
Metadherin is overexpressed in breast cancer tissue and xenografts, with lower expression in normal breast tissue. It binds to lung vasculature—one of the common sites of breast cancer metastasis—via its C-terminal extracellular domain. Blocking this domain or inhibiting Metadherin expression has been reported to reduce breast cancer metastasis.
