Product Specific Information

This antibody binds to sequence 210-241 of tau protein.

Target Information

Tau is a neuronal microtubule-associated protein found predominantly on axons. Its primary function is to promote tubulin polymerization and stabilize microtubules. The C-terminus binds axonal microtubules while the N-terminus binds neural plasma membrane components, suggesting that tau acts as a linker between both structures.

Axonal polarity is determined by TAU/MAPT localization near the centrosome. Short isoforms contribute to cytoskeletal plasticity, while longer isoforms aid in stabilization. In its hyper-phosphorylated form, Tau is the major component of paired helical filaments (PHF), which form neurofibrillary lesions in Alzheimer’s disease (AD) brains. Hyper-phosphorylation impairs Tau’s microtubule-binding ability, leading to microtubule destabilization and neuronal degeneration.

Tau phosphorylation is mediated by kinases such as GSK-3β, PKA, cdk5, and casein kinase II. Hyper-phosphorylated Tau is also found in neurofibrillary lesions associated with other CNS disorders, including Pick’s disease, frontotemporal dementia, cortico-basal degeneration, and progressive supranuclear palsy.
