Product Specific Information

This GMPR9 monoclonal antibody reacts with human and mouse Sox9, a member of the Sry HMG-box (Sox) gene family of transcription factors. Sox9 contains a high mobility group (HMG-box) DNA-binding domain that recognizes the sequence AGAACAATGG and, with co-factors, bends DNA to facilitate chromatin remodeling and transcriptional regulation. Sox9 is expressed in multiple tissues during development, contributing to cartilage formation, sex determination, neural crest formation, and retinal development. It regulates genes such as sox5, sox6, sox10, collagen type 2 alpha, collagen 9A1, CEACAM1, and snail2. Dysregulation of Sox9 is associated with craniofacial defects, cleft palate, sex reversal, and various cancers (melanoma, lung adenocarcinoma, breast cancer). Sox9 is also expressed in adult stem cells of the liver, pancreas, and intestine.

Applications Reported:

  • Immunohistochemical staining of formalin-fixed paraffin-embedded tissues
  • Microscopy
  • Immunocytochemistry

Applications Tested:

  • Tested by immunocytochemistry of fixed and permeabilized cells
  • Recommended use ≤10 µg/mL (optimize by titration)

Target Information

SOX9 plays a critical role in sex determination and Sertoli cell differentiation. It regulates genes involved in chondrogenesis and acts as a transcription factor for these genes. Gene translocation can cause campomelic dysplasia. SOX9 contributes to testis formation, neuron-glia switch in spinal cord development, and prostate epithelium differentiation. In prostate cancer, SOX9 function is altered to promote tumor growth and invasion. It also regulates neural crest precursor formation and melanin production. Expression has been observed in adult testis, adult heart, and fetal brain.
