Product Specific Information

The monoclonal antibody 1A4 recognizes human, mouse, and rat alpha-smooth muscle actin (α-SMA), a 42 kDa protein and major component of the cytoskeletal structural network. This antibody is specific for the alpha form of muscle actin expressed by smooth muscle cells of blood vessels, myofibroblasts, and myoepithelial cells. It is also expressed in various tissue parenchyma and stroma.

Alpha-smooth muscle actin is useful for identifying leiomyomas, leiomyosarcomas, pleomorphic adenomas, and serves as a prognostic marker for basal cell carcinoma.

Applications Reported:

• Immunohistochemical staining of formalin-fixed paraffin-embedded tissue sections
• Immunocytochemistry

Applications Tested:

• IHC (P): ≤1 µg/mL using low pH antigen retrieval
• ICC: ≤1 µg/mL using methanol-fixed cells
  Careful titration is recommended for optimal performance.

Target Information

Smooth Muscle Actin belongs to the actin family, which plays a critical role in cell motility, structure, and integrity. Alpha, beta, and gamma actin isoforms are identified, with alpha actin being a major constituent of the contractile apparatus. Smooth muscle actin (α-actin) is found in skeletal muscle and smooth muscle vessel walls, gut wall, myometrium, and myoepithelial cells.

Defects in the smooth muscle actin gene can cause familial thoracic aortic aneurysm type 6. Alpha-smooth muscle actin is abundant in vascular and visceral smooth muscle cells and appears in fibroblastic stress fibers during wound healing and fibrocontractive diseases.
