Product Specific Information
The monoclonal antibody 7F9 (VIIF9) recognizes human, mouse, rat, and avian vinculin and its alternatively spliced isoform, metavinculin. Vinculin is a cytoskeletal protein associated with cell-cell and cell-matrix junctions, involved in anchoring F-actin to the membrane and regulating E-cadherin expression. It binds to talin, paxillin, and alpha-actinin. Dysregulation of vinculin affects cell adhesion, migration, and growth, promoting cancer invasion and metastasis.
Applications Reported:
Used in immunohistochemical staining of frozen and formalin-fixed paraffin-embedded tissues, microscopy, and immunocytochemistry.
Applications Tested:
Validated by immunocytochemistry of formaldehyde-fixed and permeabilized cells, usable at ≤20 µg/mL. Recommended titration for optimal performance.

Target Information
Vinculin (VCL) is a cytoskeletal protein found at cell-cell and cell-extracellular matrix adherens-type junctions. It anchors F-actin to the membrane through interactions with talin, alpha-actinin, and integrin. This linkage forms part of adhesion plaque assembly, crucial for cytoskeletal integrity and cell adhesion. Mutations in the VCL gene are associated with cardiomyopathy (dilated 1W or familial hypertrophic 15).
