Description:
The FF3399 monoclonal antibody recognizes human aryl hydrocarbon receptor (AHR), a ligand-activated transcription factor mediating toxic responses to environmental contaminants such as PCBs and TCDD. AHR also binds naturally occurring compounds from fruits, vegetables, and metabolites. It is found in the cytoplasm complexed with HSP90, p23, and XAP2/AIP/ARA9. Upon ligand binding, AHR translocates to the nucleus, associates with ARNT, and binds to the consensus DNA sequence GCGTG to regulate genes such as CYP1A1. AHR is expressed in many tissues, with the highest levels in the liver, and is involved in Treg and Th17 cell regulation.

Fluorochrome-conjugated FF3399 is recommended for intracellular flow cytometry.

Applications Tested:
Tested in IHC (FFPE), ICC/IF on fixed and permeabilized human cells, and WB. Optimal concentration ≤20 µg/mL; titration recommended for best results.

Target Information

AHR (Ah Receptor) is part of a protein family including ARNT (HIF-1 Beta) and Drosophila PER/SIM proteins. It contains an N-terminal PAS domain (~200 aa). AHR binds specific DNA enhancer sequences to initiate transcription of the cytochrome P-450 (CYP1A1) gene. It functions as a ligand-activated transcription factor regulating responses to aromatic hydrocarbons and xenobiotic-metabolizing enzymes. Ligands include polycyclic and halogenated aromatic compounds such as TCDD. In inactive cells, AHR is cytoplasmic and complexed with HSP90; upon ligand binding, it translocates to the nucleus and forms a heterodimer with ARNT. AHR is associated with conditions such as eosinophilic fasciitis and seborrheic dermatitis.
