Description:
The monoclonal antibody 10C2 recognizes human nestin (residues 1464–1614). Nestin is a 220–240 kDa Class VI intermediate filament protein expressed in stem cells of the developing nervous system. Expression is also observed in pancreatic islet and mesenchymal precursors as well as cells with angiogenic potential. Upon differentiation, nestin-containing filaments are replaced by cell type-specific intermediate filament proteins such as GFAP. Expression has been identified in various cancerous cells including glioblastomas and pancreatic tumors. No cross-reactivity with rodent nestins has been observed using the 10C2 antibody.

Applications Reported:
This antibody is suitable for immunocytochemistry, microscopy, western blotting, immunohistochemical staining of frozen and paraffin-embedded tissue sections, and flow cytometry.

Applications Tested:

• Tested by western blot on SDS-reduced and non-reduced lysates from U251 cells (≤5 µg/mL).
• Tested by immunocytochemistry on formaldehyde-fixed and permeabilized SKNSH cells (≤5 µg/mL).
• Recommended to titrate for optimal performance in specific assays.

Purity: Greater than 90%, determined by SDS-PAGE.
Aggregation: Less than 10%, determined by HPLC.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

Nestin is a Class VI intermediate filament abundantly produced in the developing central nervous system (CNS) and in early embryonic neuroepithelial stem cells. It serves as a predominant marker for stem/progenitor cells, glioma cells, and tumor endothelial cells. Nestin is also a superior angiogenic marker for evaluating neovascularity in tumors. Highly specific antibodies to human Nestin aid in characterizing progenitor cells differentiating into distinct lineages, enhancing therapeutic potential of neural stem/progenitor cells in CNS disease or injury, and identifying neuroepithelial tumor cells. Nestin expression has been observed in pancreatic hematopoietic stem cell populations and is considered a novel angiogenesis marker in proliferating endothelial cells of colorectal cancer tissue.
