Preparation:
Produced from sera of rabbits immunized with highly pure Recombinant Human sCD22. Anti-Human sCD22-specific antibody was purified by affinity chromatography employing an immobilized Human sCD22 matrix.

Sandwich ELISA:
To detect Human sCD22 by sandwich ELISA (using 100 µL/well antibody solution), a concentration of 0.5–2.0 µg/mL is required. This antigen affinity purified antibody, in conjunction with PeproTech Biotinylated Anti-Human sCD22 (500-P227Bt) as a detection antibody, allows detection of at least 0.2–0.4 ng/well of Recombinant Human sCD22.

Western Blot:
To detect Human sCD22 by Western Blot analysis, this antibody can be used at a concentration of 0.1–0.2 µg/mL. With compatible secondary reagents, the detection limit for Recombinant Human sCD22 is 1.5–3.0 ng/lane under reducing or non-reducing conditions.

Target Information

CD22 (BL-CAM) is a type 1 integral membrane glycoprotein (130–140 kDa) expressed in both the cytoplasm and membrane of B-lymphocytes. It appears early in B-cell differentiation, similar to CD19, and its membrane expression is limited to mature B cells (CD22+) and plasma cells (CD22−). This makes CD22 useful in phenotyping mature leukemias. It is strongly expressed in hairy cell leukemia and positive in diffuse large B-cell lymphoma and nodular lymphocyte predominance Hodgkin’s lymphoma, but negative in classical Hodgkin’s lymphoma.
CD22 preferentially binds to α2,6-linked sialic acid, and its recognition site can be masked by cis interactions. CD22 is involved in regulation of B-cell receptor signaling through ligand-induced tyrosine phosphorylation and interactions with Src family kinases and cytoplasmic phosphatases.
