Preparation:
Produced from sera of rabbits immunized with highly pure Recombinant Human sCD22. Anti-Human sCD22-specific antibody was purified by affinity chromatography and then biotinylated.

Sandwich ELISA:
To detect hCD22 by sandwich ELISA (using 100 µL/well antibody solution), a concentration of 0.25–1.0 µg/mL is required. This biotinylated polyclonal antibody, in conjunction with PeproTech Polyclonal Anti-Human CD22 (500-P227) as a capture antibody, allows detection of at least 0.2–0.4 ng/well of Recombinant hCD22.

Western Blot:
To detect hCD22 by Western Blot, use at 0.1–0.2 µg/mL. With compatible secondary reagents, detection limit for Recombinant hCD22 is 1.5–3.0 ng/lane under reducing or non-reducing conditions.

Target Information

CD22 (BL-CAM) is a type 1 integral membrane glycoprotein (130–140 kDa) expressed in cytoplasm and membrane of B-lymphocytes. CD22 appears early in B-cell differentiation, similar to CD19, and is limited to mature B cells (CD22+) and plasma cells (CD22−). Useful for phenotyping mature leukemias and strongly expressed in hairy cell leukemia.
CD22 preferentially binds α2,6-linked sialic acid. Its recognition site may be masked by cis interactions with sialic acids on the same surface. Ligand-induced tyrosine phosphorylation regulates B-cell receptor signaling, acting as both positive and inhibitory receptor through interactions with Src kinases and phosphatases.
CD22 is also positive in diffuse large B-cell lymphoma and nodular lymphocyte predominance Hodgkin’s lymphoma, but negative in classical Hodgkin’s lymphoma.
