Preparation:
Produced from sera of rabbits immunized with highly pure Recombinant Human sCD22. Anti-Human sCD22-specific antibody was purified by affinity chromatography and then biotinylated.

Sandwich ELISA:
To detect hCD22 by sandwich ELISA (using 100 µL/well antibody solution), a concentration of 0.25–1.0 µg/mL of this antibody is required. This biotinylated polyclonal antibody, in conjunction with PeproTech Polyclonal Anti-Human CD22 (500-P227) as a capture antibody, allows detection of at least 0.2–0.4 ng/well of Recombinant hCD22.

Western Blot:
To detect hCD22 by Western Blot analysis, use at 0.1–0.2 µg/mL. In conjunction with compatible secondary reagents, the detection limit for Recombinant hCD22 is 1.5–3.0 ng/lane under either reducing or non-reducing conditions.

Target Information

CD22 (BL-CAM) is a type I integral membrane glycoprotein (130–140 kDa) expressed in both the cytoplasm and membrane of B-lymphocytes. It appears early in B-cell differentiation, similar to CD19. Unlike other B-cell markers, CD22 membrane expression is limited to the late differentiation stages between mature B cells (CD22⁺) and plasma cells (CD22⁻), making it useful in phenotyping mature leukemias. CD22 is strongly expressed in hairy cell leukemia, and positive in diffuse large B-cell lymphoma and nodular lymphocyte predominance Hodgkin’s lymphoma, but negative in classical Hodgkin’s lymphoma. It binds preferentially to α2,6-linked sialic acid and is involved in regulation of B-cell receptor signaling through interactions with Src family kinases and cytoplasmic phosphatases.
