Product Specific Information

The mShad150 monoclonal antibody reacts with mouse CD150, a ~70 kDa transmembrane glycoprotein also known as Signaling Lymphocyte Activation Molecule (SLAM). CD150 is expressed by T cells (particularly Th1) and B cells, with expression rapidly upregulated upon activation. Immature thymocytes and dendritic cells also express this antigen.

Signaling through SLAM in T cells induces proliferation and enhances interferon-gamma response. SLAM also plays a role in adhesion between T cells and antigen-presenting cells. The mShad150 antibody stains the CD150⁺CD244⁻CD48⁻CD41⁻ population of pluripotent hematopoietic stem cells.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes at ≤0.5 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Optimize titration for best performance.

Fluorochrome Information:
Super Bright 645 is a tandem dye excited by the violet laser (405 nm) with emission at 645 nm. Recommended filter: 660/20 bandpass. Ensure instrument compatibility.

Staining Recommendations:
When using multiple Super Bright-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity:
Protect from light to prevent photo-induced oxidation.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal signal loss.

Target Information

CD150 (SLAM, Signaling Lymphocyte Activation Molecule)
CD150 belongs to the immunoglobulin gene superfamily and is involved in T cell stimulation. It is constitutively expressed on peripheral blood memory T cells, T cell clones, immature thymocytes, and some B cells, and is rapidly induced on naive T cells after activation. Activated B cells express membrane-bound, soluble, and cytoplasmic isoforms of SLAM.

SLAM-SLAM binding enhances B cell activation and differentiation. The extracellular domain of CD150 serves as the receptor for measles virus and acts as a co-activator for T and B cells. CD150 interacts with SH2D1A and PTPN11 via its cytoplasmic domain. Mutations in the CD150 gene may be associated with X-linked lymphoproliferative disease (XLP).
