Product Specific Information

Description:
The eBioRDR5 monoclonal antibody reacts with human CD127 (Interleukin-7 Receptor alpha). CD127 complexes with CD132 (common gamma chain, γc) to form the IL-7 receptor (IL-7R). CD127 is a type I glycoprotein (~75–80 kDa) expressed by immature B cells, thymocytes, and most mature T cells, with transient down-regulation upon activation. IL-7 binding triggers signal transduction via the Jak/STAT pathway, essential for lymphocyte development and T-cell homeostasis.

CD127 expression is down-regulated on CD4⁺CD25⁺ regulatory T cells (Tregs). Co-expression of CD4 and CD25 alone may not define suppressive cells, but CD127 down-regulation enriches for functional Tregs expressing Foxp3. Binding of eBioRDR5 to PBMCs is blocked by recombinant human IL-7 (Product #14-1079-80).

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated for flow cytometry of normal human PBMCs at 5 µL (0.25 µg)/test (100 µL final volume, 10⁵–10⁸ cells/test).

Fluorochrome Information:
Super Bright 645 is a tandem dye excited by the violet laser (405 nm) and emits at 645 nm. Use a 660/20 bandpass filter. Ensure instrument compatibility.

Staining Recommendations:
When using multiple Super Bright conjugates, use Super Bright Complete Staining Buffer (Product #SB-4401) to minimize non-specific interactions.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) with minimal impact on brightness.

Target Information

CD127 (Interleukin-7 Receptor α, IL7Rα) is a glycoprotein regulating lymphopoiesis. The active receptor is a heterodimer of α and γ chains. The γc chain activates signal transduction, while the α chain mediates specific signaling. CD127 promotes proliferation of precursor B cells, thymocytes, T-cell progenitors, and mature CD4⁺/CD8⁺ T cells. Dysregulation is linked to severe combined immunodeficiency (SCID) variants.
