Product Specific Information

The 104D2 monoclonal antibody reacts with human CD117 (c-Kit), also known as Steel factor receptor and stem cell factor receptor. This 145 kDa tyrosine kinase receptor is expressed by hematopoietic progenitor cells and mast cells. The interaction of c-Kit and Steel factor promotes proliferation and differentiation of hematopoietic progenitors and mast cells. CD117 is also expressed by melanocytes and is involved in their signaling and activation.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested by flow cytometry using normal human peripheral blood cells. Use 5 µL (0.25 µg) per test (100 µL final volume).

Super Bright 645 is a tandem dye excited by the violet laser (405 nm) and emits at 645 nm. Use a 660/20 bandpass filter. Ensure instrument compatibility.

When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity: Protect from light to prevent photo-induced oxidation.
Fixation: Samples can be stored up to 3 days in the dark at 4°C using IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) with minimal impact on brightness and FRET efficiency.

Target Information

KIT (c-KIT) is a proto-oncogene and type 3 transmembrane receptor for MGF (mast cell growth factor, or stem cell factor). KIT regulates growth and activation of hematopoietic and non-hematopoietic cells. Mutations in KIT are linked to gastrointestinal stromal tumors, mast cell disease, acute myelogenous leukemia, and piebaldism. The D816V mutation in KIT is a therapeutic target for neoplastic mast cell growth.

c-Kit is expressed in hematopoietic stem cells, germ cells, mast cells, and gastrointestinal Cajal cells. Upon SCF binding, c-Kit dimerizes, activating autophosphorylation and downstream signaling pathways (Ras/ERK, PI3K, PLC-γ, JAK/STAT), essential for hematopoiesis, stem cell maintenance, and gametogenesis.
