Product Specific Information

The A7R34 monoclonal antibody reacts with mouse CD127, the high affinity alpha subunit of the IL-7 receptor.
CD127 is expressed by immature B cells, double-negative thymocytes (CD4-CD8-), single-positive thymocytes (CD4+ or CD8+), but not double-positive (CD4+CD8+) thymocytes. Mature peripheral T cells express CD127 at low levels.

A7R34 inhibits IL-7 binding to its receptor and is used in vivo and in vitro to study IL-7’s role in T/B cell development and activation. Binding of A7R34 blocks SB/199 antibody binding to mouse CD127.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on mouse splenocytes; ≤1 µg/test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Optimize by titration.

Super Bright 645 is a tandem dye excited by violet laser (405 nm) and emits at 645 nm. Recommended filter: 660/20 bandpass. Ensure instrument compatibility.

When using multiple Super Bright dyes, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light sensitivity: Protect vial and samples from light.
Fixation: Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) with minimal impact on brightness or FRET efficiency. Clone-specific performance should be verified empirically.

Target Information

CD127 (Interleukin-7 receptor alpha, IL7Rα) is a glycoprotein regulating lymphopoiesis. The active receptor is an alpha/gamma chain heterodimer.

  • Gamma(c) chain activates IL7R signal transduction.
  • Alpha chain mediates specific signaling.
    CD127 promotes proliferation of precursor B cells, thymocytes, T cell progenitors, and mature CD4+/CD8+ T cells.
    Dysfunction is associated with severe combined immunodeficiency (SCID) forms.
