Product Specific Information

The Canine Fc Receptor Binding Inhibitor is used to inhibit non-specific Fc receptor (FcR)-mediated binding of rat monoclonal antibodies during flow cytometric analysis of canine tissue.
FcR are expressed in multiple cell lineages, with high levels in myeloid cells, granulocytes, and B cells. Their biological functions—endocytosis, phagocytosis, and antigen presentation—are triggered upon host-immunoglobulin binding.

eBioscience testing indicates that rat IgG2b isotype binding is blocked by the Canine Fc Receptor Binding Inhibitor.

Applications Reported:
Canine Fc Receptor Binding Inhibitor Purified has been reported for use in flow cytometric analysis.

Usage Instructions:
To inhibit non-specific binding of rat monoclonal antibodies, add the inhibitor to samples and incubate on ice for 20 minutes. Without washing, proceed to stain with the primary antibody.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis on canine peripheral blood cells.
Recommended usage: 5 µL (10 µg) per test.
A test is defined as the antibody amount (µg) that stains a cell sample in a final volume of 100 µL. Cell number may range from 10⁵ to 10⁸ cells/test.

Purity: Greater than 90%, as determined by SDS-PAGE.
Aggregation: Less than 10%, as determined by HPLC.
Filtration: 0.2 µm post-manufacturing filtered.


Target Information

Four classes of Fcγ receptors have been identified: FcγRI (CD64), FcγRII (CD32), and FcγRIII (CD16).
They are expressed at varying levels across cell lineages, including high expression in myeloid, granulocyte, and B cells.
FcγR biological functions—endocytosis, phagocytosis, and antigen presentation—are elicited upon host-immunoglobulin binding.
Binding of monoclonal antibodies to FcγRs varies depending on antibody isotype and even among antibodies of the same isotype.
