Specificity Comments:
이 항체는 태반 3기에서 발생하는 70kDa 막결합형 isozyme (Regan 및 Nagao type) PLAP에 특이적으로 반응하며, 다른 알칼리성 인산화효소 isozyme과 교차반응하지 않습니다. Germ cell tumor와 반응하며, 다른 신생물과 구별할 수 있습니다. 유방, 위장, 전립선, 요로암 등의 체세포성 종양에서도 PLAP 항체와 면역반응을 보일 수 있습니다. Anti-PLAP 양성 및 anti-keratin 음성은 seminoma를 carcinoma와 구분하는 데 도움이 됩니다. Germ cell tumor는 일반적으로 anti-keratin 양성이지만 anti-EMA에는 반응하지 않으며, 대부분의 carcinoma는 anti-EMA에 반응합니다. Anti-PLAP은 임신성 융모성 질환 진단에도 유용합니다.
Target Information
Placental Alkaline Phosphatase는 염증성 질환 조절에 중요한 역할을 합니다. 이 효소에는 intestinal, placental, placental-like, liver/bone/kidney 등 4가지 관련 형태가 존재합니다. PLAP는 태반의 3기에서 나타나는 막결합형 isoenzyme으로, 고환 germ cell tumor 식별에 유용합니다. Germ cell tumor와 달리 PLAP 양성 체세포 종양은 epithelial membrane antigen (EMA)을 발현합니다. PLAP의 기능은 matrix mineralization으로 제안되었으며, 기능적 효소가 결핍된 생쥐에서도 정상적인 골격 발달을 보입니다. PLAP는 hypophosphatasia(고칼슘혈증 및 골격 이상을 특징으로 하는 질환)와 직접적으로 관련이 있으며, 돌연변이에 따라 발병 시기와 증상이 달라질 수 있습니다.
