• For IHC (PFA-fixed): Incubate antibody for 30 minutes at room temperature (RT).
• Formalin-fixed tissues require heating in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at RT for 20 minutes.

Target Information

Placental Alkaline Phosphatase (PLAP) plays a key role in inflammatory disease regulation. There are four related alkaline phosphatases: intestinal, placental, placental-like, and liver/bone/kidney types.
PLAP is a membrane-bound isoenzyme (Regan and Nagao types) expressed in the placenta during the third trimester. It is useful for identifying testicular germ cell tumors.
Unlike germ cell tumors, PLAP-positive somatic cell tumors uniformly express epithelial membrane antigen (EMA).
PLAP is proposed to function in matrix mineralization, although mice lacking this enzyme show normal skeletal development.
Defects in PLAP are associated with hypophosphatasia, a disorder involving hypercalcemia and skeletal abnormalities, with severity depending on the specific mutation.
