Product Specific Information
The OKT8 monoclonal antibody reacts with the human CD8a molecule, an approximately 32–34 kDa cell surface receptor expressed as a heterodimer (CD8αβ) or homodimer (CD8αα). It is expressed on most thymocytes and subsets of mature T cells and NK cells. CD8 binds MHC class I and associates with protein tyrosine kinase p56lck, playing a role in T-cell development and activation.
Preliminary testing shows that OKT8 does not compete with clones RPA-T8 or HIT8a for binding to human peripheral blood leukocytes, indicating distinct epitopes.
Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated for flow cytometry of normal human peripheral blood cells.
Use at 20 µL (0.06 µg)/test. A test is defined as the amount of antibody staining a cell sample (10⁵–10⁸ cells) in 100 µL final volume.
Excitation: 488 nm; Emission: 519 nm; Laser: Blue.
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
CD8 (Cluster of Differentiation 8) is a type I transmembrane glycoprotein of the immunoglobulin receptor family involved in signal transduction and T cell differentiation. It functions as a co-receptor with TCR for MHC-I molecules and is primarily expressed on cytotoxic T cells.
CD8α chain is essential for MHC-I binding and also expressed on subsets of NK cells, monocytes, and dendritic cells. Engagement with MHC-I/peptide complexes triggers Lck recruitment, leading to cytokine production and CTL activation. Activated CTLs are key in pathogen and tumor clearance. Differentiation of naïve CD8⁺ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
