Product Specific Information

The 4B12 monoclonal antibody reacts with mouse CCR7 (CD197), also known as EBI-1. CCR7 is a chemokine receptor for CCL19 (ELC, MIP3β) and CCL21 (SLC, MIP2β), mediating lymphocyte migration to secondary lymphoid organs (SLO). It regulates T cell trafficking, dendritic cell localization, and B cell positioning during immune responses.

Human CCR7 is a marker for central memory T cells (TCM) and plays a role in thymocyte development. Deficiency leads to altered thymic architecture and impaired T cell maturation.

For optimal visualization, staining at 37°C yields brighter signals and better resolution than 2–8°C. Co-staining with other antigens is generally unaffected but should be validated experimentally.

  1. Prepare cell suspension and block FcγIII/II receptors using 5 µg/million cells anti-mouse CD16/32 for 15 min on ice.
  2. Add 1 µg/million cells of 4B12 antibody and incubate at 37°C or 2–8°C for 30 min.
  3. Wash once with Flow Cytometry Staining Buffer and decant.
  4. Analyze by flow cytometry or proceed with secondary staining.

Note: Co-staining with CCR7 ligand CCL19-Fc may be hindered due to steric interference. 4B12 binding can block CCL19-Fc detection, but not vice versa.


Target Information

CCR7 is a G protein-coupled receptor induced by Epstein-Barr virus (EBV) and expressed in blood, bone marrow, lymph node, and intestine. It regulates migration of memory T cells and dendritic cell maturation. Ligands include CCL19 (ECL) and CCL21. CCR family members (CCR1–CCR5) play key roles in T cell chemotaxis and phagocyte migration to inflammation sites.
