Product Specific Information

• Source: Monoclonal antibody obtained by fusing the P3-X63-Ag8-U1 mouse myeloma cell-line with spleen cells of Donryu rat after immunization with mouse endoderm cell line PSA5-E.
• Specificity: Reacts specifically with mouse P-cadherin and inhibits P-cadherin-dependent cell-cell contact.
• Cross Reactivity: Does not react with human or chicken P-cadherin.
• Storage: Store at 4°C. Product does not contain preservative. Stock solution (2.0 mg/mL) can be stored at -20°C for 1 year, or at 4°C for 6 months with 0.1% sodium azide added. Avoid repeated freeze-thaw cycles.
• Reconstitution: Dissolve lyophilized antibody in 50 µL distilled water (final concentration: 2.0 mg/mL). For short-term use, may dissolve directly in 500 µL or more of dilution solution. Maintain minimum concentration of 2.0 mg/mL for stability. Reconstituted solution should contain 0.1% sodium azide when stored at 4°C.
• Buffer Note: Ca²⁺ should be included in buffers to stabilize cadherin antigen.

Target Information

P-cadherin (Cadherin-3) is a Type 1 cadherin protein belonging to the cadherin superfamily. It is a single-pass transmembrane glycoprotein with five extracellular cadherin repeats and an intracellular domain binding p120-catenin and beta-catenin. Cadherins mediate calcium-dependent cell-cell adhesion and are involved in development, wound healing, signaling, growth, and differentiation.

P-cadherin is expressed in fetal tissues and adult structures such as epidermis, breast, prostate, ovary, cervix, hair follicle, and corneal endothelium. It is also found in embryonic stem cells and mammary and hair follicle stem cells. Overexpression correlates with poor prognosis in several carcinomas (breast, prostate, ovary, colon, stomach). Mutations in CDH3 are linked to congenital hypotrichosis with juvenile macular dystrophy.
