Product Specific Information

The 3D12 monoclonal antibody reacts with human CCR7 (CD197), also known as EBI-1. CCR7 is a G-protein-coupled chemokine receptor involved in cell migration and immune cell trafficking. It binds to CCL19 and CCL21 and is expressed on T cells and dendritic cells (DCs).
During DC maturation, CCR7 expression increases and plays roles in chemotaxis to lymph nodes, endocytosis rate, survival, and maturation. Expression can be downregulated upon ligand binding.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on normal human peripheral blood cells at ≤ 1 µg/test. A test is defined as the antibody amount for staining a 100 µL cell sample (10⁵–10⁸ cells/test). Optimize by titration for best performance.
Recommended incubation: ≥ 45 minutes at 2–8°C for optimal staining.

Target Information

CCR7 belongs to the G protein-coupled receptor family and is induced by Epstein-Barr virus (EBV). It is expressed in blood, bone marrow, lymph node, and intestine, particularly in activated B and T lymphocytes. CCR7 regulates memory T cell migration and dendritic cell maturation. Its ligands include CCL19 and CCL21. Other C–C chemokine receptors include CCR1–CCR5 and the Duffy antigen, which are key in T cell chemotaxis and phagocyte migration during inflammation.
