Product Specific Information

The monoclonal antibody CXCR3-173 recognizes mouse CD183 (CXCR3), a seven-transmembrane G-protein-linked chemokine receptor binding three ligands: CXCL9 (Mig), CXCL10 (IP-10), and CXCL11 (ITAC).
CD183 plays a role in CD4 T cell responses to grafts, and CXCR3 knockout mice show compromised allograft rejection. Expression occurs on NK cells, T lymphocyte subsets, Tregs, and is preferentially found on Th1-polarized cells.

The antibody has been shown to affect chemotaxis in response to ligand; ligand presence eliminates staining. In vivo addition delays cardiac and pancreatic islet allograft rejection.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested by flow cytometric analysis of mouse splenocytes at ≤0.25 µg/test.
A test is defined as the amount of antibody staining a cell sample in 100 µL final volume.
Cell number may range from 10⁵–10⁸ cells/test and should be titrated for optimal performance.

Filtration: 0.2 µm post-manufacturing filtered.


Target Information

This gene encodes a G protein-coupled receptor selective for chemokines IP10, Mig, and I-TAC, belonging to the CXC chemokine subfamily.
Binding induces cellular responses related to leukocyte traffic, integrin activation, cytoskeletal changes, and chemotactic migration.
Inhibition by Bordetella pertussis toxin indicates coupling with Gi-subclass G proteins.

Signal transduction may involve enzymes similar to other chemokine receptors.
Chemokine-induced desensitization leads to rapid, short-duration responses followed by receptor recycling.
Prominent expression occurs in effector/memory T cells and inflamed tissues, suggesting involvement in inflammatory cell recruitment.
This receptor is a potential therapeutic target for small-molecule antagonists in inflammatory diseases.
Multiple transcript variants encoding different isoforms have been identified.
