Product Specific Information

Description:
The 1D1 monoclonal antibody reacts with rat Bax (~21 kDa) and does not react with human or mouse Bax. It shows no cross-reactivity with Bcl-2 or Bcl-xL proteins. Bcl-2, Bax, and Bcl-xL are members of a protein family that regulates apoptosis. While Bcl-2 and Bcl-xL inhibit apoptosis, overexpression of Bax promotes programmed cell death. The immunogen was a rat Bax peptide sequence corresponding to amino acids 3–16.

Applications Reported:
The 1D1 antibody is reported for use in immunoprecipitation and Western blotting (WB).

Applications Tested:

  • Western Blot: ~1 µg/mL
  • Immunoprecipitation: ~1–10 µg/mL
    It is recommended to titrate the reagent for optimal performance.

Purity: Greater than 90%, determined by SDS-PAGE.
Aggregation: Less than 10%, determined by HPLC.
Filtration: 0.2 µm post-manufacturing filtered.


Target Information

BAX is a member of the Bcl-2 family and plays an important role in regulating apoptosis. While Bcl-2 is anti-apoptotic, BAX promotes apoptosis. Regulation involves homo- and heterodimerization of BAX and Bcl-2 isoforms. The Bax gene encodes isoforms including Bax alpha (21 kDa) and Bax beta (24 kDa), both containing BH1, BH2, and BH3 domains. Bax beta has a unique C-terminus lacking a transmembrane domain.
Bcl-2 is also expressed in multiple isoforms; Bcl-2 beta (22 kDa) differs from alpha (26 kDa) in its C-terminus and 3′ UTR.
Overexpression of BAX promotes cell death, and it can form homodimers or heterodimers with other BCL-2-related proteins.
