Product Specific Information
Description:
The 9E10 antibody reacts with human c-Myc p67. It was raised against a synthetic peptide corresponding to amino acids 408–439 within the carboxy-terminal domain of human c-Myc. This antibody can detect Myc-tagged proteins.
The transcription factor c-Myc is a proto-oncogene involved in cell cycle regulation, metabolism, apoptosis, differentiation, cell adhesion, and tumorigenesis. In normal cells, c-Myc expression is tightly regulated, but in many human cancers, it is deregulated. c-Myc also plays a key role in apoptosis, particularly in connection with the CD95/Fas death receptor pathway.
Applications Reported:
This 9E10 antibody has been reported for use in immunoprecipitation, western blotting, and immunohistochemistry of formalin-fixed paraffin-embedded tissue sections.
Applications Tested:
- Tested by IHC using low or high pH antigen retrieval at ≤5 µg/mL.
- Tested by western blot at ≤5 µg/mL.
It is recommended to titrate the antibody for optimal performance in each assay.
Purity: >90%, as determined by SDS-PAGE
Aggregation: <10%, as determined by HPLC
Filtration: 0.2 µm post-manufacturing filtered
Target Information
The c-Myc protein is a 62 kDa transcription factor encoded by the c-Myc gene on human chromosome 8q24. It is commonly activated in various tumor cells and plays an important role in proliferation, differentiation, apoptosis, and cell cycle progression.
Phosphorylation at Thr58/Ser62, mediated by kinases such as GSK3, CDK, ERK2, and JNK, regulates c-Myc function. c-Myc is essential for vasculogenesis and angiogenesis in tumor development.
The c-myc oncogene is involved in normal cellular proliferation and differentiation, while deregulated expression induces apoptosis. Antibodies against c-Myc epitopes recognize overexpressed proteins containing Myc epitope tags.
Amplification of c-Myc occurs in several human tumors, including lung, breast, and colon carcinomas. Translocation of the c-myc locus on chromosome 8 to immunoglobulin loci is characteristic of Burkitt’s lymphoma and other B-cell conditions. Aberrant expression is found in tumors of multiple origins, including colorectal, gastric, hepatic, ovarian, and hematopoietic neoplasms.
