EGFR은 세포막 수용체로, EGF 및 TGF-α와 같은 리간드 결합 시 이합체화 및 자가 인산화가 일어나며, Ras-ERK, PI3K-Akt, Jak-STAT, PKC 등 다양한 신호 경로를 활성화합니다.
EGFR의 Y1086 인산화는 GRB2 단백질 결합을 유도하여 MAPK 경로를 활성화합니다. 활성화된 EGFR은 내포화 후 분해되거나 재활용됩니다.
EGFR 유전자 변이는 폐암 등 다양한 암과 관련이 있으며, 과발현은 예후 불량과 연관됩니다. 두경부암, 뇌종양, 방광암, 위암, 유방암, 폐암, 자궁내막암, 자궁경부암 등 다양한 종양에서 EGFR 과발현이 관찰됩니다.
