Product Specific Information
Heat-mediated antigen retrieval is recommended prior to staining, using a 10 mM citrate buffer (pH 6.0) for 10 minutes followed by cooling at room temperature for 20 minutes.
After antigen retrieval, incubate samples with the primary antibody for 30 minutes at room temperature.
A suggested positive control is breast carcinoma tissue.
Target Information
The ID (Inhibitor of DNA binding) family of basic helix-loop-helix (bHLH) proteins includes ID1, ID2, ID3, and ID4. These proteins are widely expressed and form dimers with class A and B HLH proteins.
Due to the absence of the basic region, ID proteins cannot bind DNA directly and act as negative regulators of bHLH-mediated transcription.
ID1 inhibits DNA binding of E12 and MyoD, thereby suppressing muscle-specific gene expression. During muscle cell differentiation, ID protein levels decrease, allowing E12/E47 to interact with MyoD and myogenin to activate myogenic differentiation.
Expression of ID proteins is highly dependent on growth factor signaling, and reduction of ID mRNA levels delays cell cycle reentry following growth factor stimulation.
