Target Information

Mitochondrial transcription factor A (TFAM) is a crucial protein encoded by nuclear genes but transported from the cytoplasm to mitochondria, where it plays a key role in the maintenance and expression of mitochondrial DNA (mtDNA). It regulates replication and transcription of mtDNA, ensuring proper mitochondrial function.

TFAM contains an LC3 interacting region (LIR) motif that binds autophagy proteins such as LC3, essential for the autolysosomal pathway to eliminate leaked mtDNA and limit inflammation. TFAM levels vary across tissues, influencing mitochondrial function. High TFAM levels may repress mtDNA expression in skeletal muscle, leading to OXPHOS deficiencies, while in the heart, increased mtDNA copy number maintains balance and preserves OXPHOS capacity.

Disruptions in TFAM are associated with neurodegeneration and diseases linked to mtDNA stress.
