Description:
The monoclonal antibody NVG-2 reacts with mouse CD63, also known as Lysosomal-Associated Membrane Protein 3 (LAMP-3) or tetraspanin 30 (TSPN30). CD63 belongs to the tetraspanin family characterized by four transmembrane domains. It is expressed in various hematopoietic cells (granulocytes, B lymphocytes, platelets) and non-hematopoietic cells. CD63 localizes to cell membranes, late endocytic vesicles, lysosomes, exosomes, and other granules. On the cell surface, CD63 interacts with proteins forming tetraspanin-enriched microdomains (TEM). Its high expression can indicate cell activation, making CD63 a useful activation marker for basophils, platelets, and other cells.

Applications Reported:
This NVG-2 antibody is suitable for intracellular staining followed by flow cytometric analysis.

Applications Tested:
Tested for intracellular staining of mouse peritoneal exudate cells using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00).
Refer to BestProtocols®: Protocol A for intracellular proteins.
Use ≤ 0.125 µg/test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Optimize antibody titration for best performance.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples may be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in the dark with minimal signal loss. Clone-specific performance should be verified empirically.

Target Information

CD63 (LAMP-3):
A glycoprotein of the tetraspanin family, found in late endosomes, lysosomes, and secretory vesicles. Present on plasma membranes after cell activation, CD63 serves as a basophil activation marker. It interacts with integrins, regulates phagocytosis, cell migration, and H/K-ATPase trafficking. CD63 also acts as a T-cell costimulatory molecule and can be a prognostic marker in carcinomas. Expressed on activated platelets, monocytes, macrophages, and weakly on granulocytes, B, and T cells. Identical to melanoma antigen ME491 and platelet antigen PTLGP40. Dysfunction is associated with melanoma and Hermansky-Pudlak Syndrome.
