Product Specific Information

Description:
The DREG-56 monoclonal antibody reacts with human CD62L, a 76 kDa member of the selectin family. CD62L is expressed by neutrophils, monocytes, and subsets of T, B, and NK cells. It binds glycosylated, fucosylated, sulfated sialylated glycoproteins including CD34, glycam-1, and MAdCAM-1, mediating lymphocyte rolling on activated endothelium and homing to high endothelial venules (HEV) in peripheral lymphoid tissues.

Applications Reported:
Reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Use at 5 µL (0.5 µg) per test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test (empirically determined).

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Excitation/Emission:
488–561 nm excitation; 775 nm emission.
Compatible with Blue, Green, and Yellow-Green lasers.

Filtration:
0.2 µm post-manufacturing filtered.


Target Information

The human CD62L (L-Selectin) is a 74–95 kDa glycoprotein adhesion receptor. It includes:

  • N-terminal C-type lectin binding domain
  • EGF-like domain
  • Two short consensus repeat (SCR) sequences
  • Spacer, transmembrane, and cytoplasmic regions

CD62L is constitutively expressed on most leukocytes, including lymphocytes (except some memory T cells), monocytes, and polymorphonuclear cells.
