Heat-mediated antigen retrieval is recommended prior to staining, using a 1mM EDTA buffer (pH 8.0) for 10 minutes, followed by cooling at room temperature for 20 minutes.
Following antigen retrieval, incubate samples with primary antibody for 30 minutes at room temperature.
A suggested positive control is tonsil tissue.

Target Information

Cell-mediated immune responses are initiated by T lymphocytes stimulated by peptides bound to MHC molecules on antigen-presenting cells (APC).
Activated T cells express receptors such as PD-1 (PDCD-1) that mediate inhibitory signals from APCs.
PD-1 binds two ligands, PDL-1 and PDL-2, which inhibit immune activation in the absence of danger signals like LPS.
PD-1 deficient mice exhibit hyperactivated immune systems and autoimmune diseases.
Despite its predicted molecular weight, PD-1 often migrates at a higher molecular weight in SDS-PAGE.
