Product Specific Information
Heat-mediated antigen retrieval is recommended prior to staining, using a 10 mM citrate buffer (pH 6.0) for 10 minutes, followed by cooling at room temperature for 20 minutes.
After antigen retrieval, incubate samples with the primary antibody for 10 minutes at room temperature.
A suggested positive control is breast or ovarian carcinoma tissue.
Target Information
The Notch gene family encodes transmembrane proteins with multiple cysteine-rich EGF-like repeats in the extracellular region.
Four Notch genes have been described in mammals: Notch1, Notch2, Notch3, and Notch4 (Int-3), all involved in differentiation of the nervous system and other structures.
EGF-like proteins Delta and Serrate act as ligands for Notch1.
Mature Notch proteins form heterodimeric receptors derived from cleavage of Notch pre-proteins into two subunits:
- NEC: Extracellular subunit containing multiple EGF-like repeats
- Ntm: Transmembrane subunit including intracellular region
