Heat-mediated antigen retrieval is recommended prior to staining, using a 10 mM citrate buffer (pH 6.0) for 10 minutes, followed by cooling at room temperature for 20 minutes.
Following antigen retrieval, incubate samples with primary antibody for 30 minutes at room temperature.
A suggested positive control is breast carcinoma.

Target Information

RPS6KB1 (p70S6K) is a serine/threonine kinase with two non-identical kinase catalytic domains that phosphorylate several residues of the S6 ribosomal protein.
This kinase is predominantly localized in the cytoplasm and plays a key role in growth factor–regulated cell proliferation, cell motility, immune response, and tissue repair.
p70 S6 kinase acts downstream of phosphoinositide (PI) 3-kinase, targeting the S6 ribosomal protein involved in upregulation of protein synthesis.
Its activity promotes increased protein synthesis and cell proliferation. Amplification and overexpression of p70 S6 kinase are observed in certain breast cancer cell lines.
