Specificity Comments:
Tissue transglutaminase (tTG) is a 75 kDa monomeric globular protein expressed in most cells and tissues. It localizes mainly in the cytoplasm, with some presence on the cell surface associated with the extracellular matrix (ECM). In addition to GTPase activity, tTG catalyzes posttranslational protein modification by transamidation of glutamine residues. Extracellular tTG binds and cross-links ECM proteins, contributing to ECM and basement membrane stability. High constitutive expression and accumulation of active tTG are observed in endothelial cells, platelets, vascular smooth muscle cells, and epithelial cells of the lens. Expression is low in skeletal muscle cells and neurons.

Target Information

Transglutaminase (TGase) catalyzes protein crosslinking via ε-(γ-glutamyl) lysine isopeptide bonds, requiring Ca²⁺ for activity. Eight distinct TGase isoenzymes are identified in mammals. Tissue transglutaminase (tTGase, TGase 2) comprises four domains: N-terminal β-sandwich, catalytic core, and two C-terminal β-barrel domains. tTGase is implicated in cell death, proliferation, differentiation, and receptor-mediated endocytosis. In Alzheimer’s disease, elevated tTGase activity contributes to polymerization of proteins such as Aβ peptide, β-amyloid precursor protein, and tau, forming neurofibrillary tangles.
