Target Information
Transglutaminase (TGase)는 Ca²⁺ 결합을 필요로 하는 e-(γ-glutamyl) lysine 이소펩타이드 결합을 형성하여 단백질을 교차결합하는 효소입니다. 포유류에서는 8가지 TGase 동형효소가 확인되었으며, Tissue transglutaminase (tTGase, TGase 2)는 N-말단 β-sandwich, 촉매 코어, 두 개의 C-말단 β-barrel 도메인을 포함합니다.
tTGase는 세포 사멸, 증식, 분화, 수용체 매개 엔도사이토시스에 관여할 수 있습니다. 알츠하이머병 뇌에서는 tTGase 활성 증가로 인해 Ab 펩타이드, β-amyloid 전구체 단백질, tau 단백질 등의 중합이 일어나 신경섬유 엉킴(neurofibrillary tangles)이 형성됩니다.
