Target Information
Transglutaminase (TGase)는 Ca²⁺ 결합을 필요로 하는 단백질 간의 ε-(γ-glutamyl) lysine 이소펩타이드 결합 형성을 촉매하는 효소입니다. 포유류에서는 8가지 TGase 이소효소가 확인되었으며, 그중 Tissue transglutaminase (tTGase, TGase 2)는 N-말단 β-sandwich, 촉매 코어, 2개의 C-말단 β-barrel 도메인으로 구성됩니다. tTGase는 세포사멸, 증식, 분화 및 수용체 매개 엔도사이토시스에 관여할 수 있습니다. 알츠하이머병 뇌에서는 tTGase 활성이 증가하여 Ab peptide, β-amyloid precursor protein, tau 단백질 등의 중합과 신경섬유 엉킴(neurofibrillary tangles) 형성에 기여합니다.
